Description | The B16-F10 cell line is a well-characterized murine melanoma model derived from the skin tissue of a C57BL/6 mouse. It is a subline generated through the 10th serial passage of the original B16 parent tumor line in C57BL/6 mice. This subclone is noted for its high metastatic potential in vivo and is often used in preclinical studies to evaluate the efficacy of immuno-oncology agents and the development of novel therapeutics. The B16-F10 model is used for investigations into tumor growth kinetics, blood and lymphatic vascularization, tumor and immune cell features, and the spontaneous development of metastases, thereby facilitating the development of more effective treatment strategies for melanoma??. The murine melanoma cell line B16-F10 grows adherently and displays an epithelial morphology. When implanted intradermally in C57BL/6 mice, the cells form lung colonies. The B16 tumor line, including B16-F10 cells, has been vital in studying local tumor growth and metastasis. Subculturing | Remove the old medium from the adherent cells and wash them with PBS that lacks calcium and magnesium. For T25 flasks, use 3-5 ml of PBS, and for T75 flasks, use 5-10 ml. Then, cover the cells completely with Accutase, using 1-2 ml for T25 flasks and 2.5 ml for T75 flasks. Let the cells incubate at room temperature for 8-10 minutes to detach them. After incubation, gently mix the cells with 10 ml of medium to resuspend them, then centrifuge at 300xg for 3 minutes. Discard the supernatant, resuspend the cells in fresh medium, and transfer them into new flasks that already contain fresh medium. |
